Suan-Zao-Ren Decoction improves cognitive impairment and reduces neural degeneration in APP/PS1 transgenic mice by inhibiting Aβ accumulation and neuroinflammation via the JAK2/STAT3 pathway.
The authors initiated their investigation by subjecting APP/PS1 transgenic mice to different dosages of Suan-Zao-Ren Decoction (SZRD) for a period of 4 weeks. Cognitive changes in the behavior of these mice were observed through the Morris water maze. Inside the hippocampus of the mice, the researchers utilized Nissl and Golgi-Cox staining to evaluate synaptic plasticity, and used ELISA and western blot applications to measure the proteins and expression levels related to degenerative neurological conditions.
In comparing the untreated APP/PS1 mice with the ones treated with SZRD, it was found that especially the low-dosage SZRD significantly improved cognitive impairment. This improvement was also noted alongside a decrease in neuron loss and Aβ plaque deposition, as well as an increase in hippocampus synaptic plasticity. Furthermore, SZRD demonstrated a substantial decrease in the serum and hippocampal concentrations of various inflammation indicators. Moreover, there was a noticeable reduction in the expression of certain proteins within the hippocampus of the treated mice.
