The antihypertensive activity of Tianma Gouteng Decoction is shown to be most effective when administered twice daily by mediating 15 biomarkers and influencing renin-angiotensin-aldosterone system.
In the methodology, a spontaneous hypertension rat model with live-Yang hyperactivity hypertension was established through aconite decoction application. The rats were then randomly separated into four distinct groups - a model group, and three Tianma Gouteng Decoction groups with varying frequencies of medication. Blood pressure and indexes of the renin-angiotensin-aldosterone system were measured. In addition, a metabolic profile of rat plasma samples was established using an advanced mass spectrometry technique, which was further analyzed to reveal connections between metabolic paths and hypertension.
The discussion of the results reveals that Tianma Gouteng Decoction administered twice a day shows significant blood pressure reduction in the hypertensive rat model and regulates several bio-markers associated with hypertension. Analysis of plasma metabolites identified 47 potential biomarkers in hypertensive rats, and Tianma Gouteng Decoction regulated 15 of these. The twice-daily treatment schedule demonstrated the most substantial moderating effect on these regulated biomarkers, evidencing the antihypertensive activity of Tianma Gouteng Decoction. This antihypertensive activity is linked to its influence on the renin-angiotensin-aldosterone system and regulation of various metabolisms including glycerol phospholipid, sphingomyelin, energy, and amino acid metabolisms.
